  • Introduction: The snapshot technique is a pcr-based molecular method, in which targeted primers for a specific locus are utilized in kits containing fluorescent dye-labeled ddNTPs and reads in the genetic analyzer. This method is highly practical due to the simultaneous use of multiple loci, usually between 10 and 20 dots in a single PCR reaction. It is currently being used for point mutations and polymorphisms in genetic diseases and population genetic studies.
  • Methods: primers are designed after selecting the SNPs studied. All loci are then PCR amplified in a single reaction. Next, the PCR product is purified with the EXO I - SAP enzyme, followed by the secondary PCR product purified with SBE primers and the SNAPSHOT kit. Then the reaction product is purified with the SAP enzyme. It is mixed with HIDI and LIZ 120 and read in the Genetic Analyzer. The outputs of the device are then analyzed with GENEMAPPER software.
  • Results: Four nucleotides T (red), A ( green), G ( blue), and C ( yellow ) can be seen in the normal and mutated heterozygous or homozygous.
  • Conclusion: This method is very accurate and sensitive because of its readings with the genetic analyzer and the use of the kit. Multiplexing makes this method very practical and inexpensive for several locations. The key to using this method is precision in the design of secondary primers (SBE PRIMER) and skill in laboratory procedures.
