  • Introduction: Infertility as a problem with different causes, and the number of couples suffering from infertility is increasing around the world. To this end, many approaches have been developed to help the infertile couples to have children. In vitro maturation (IVM) of oocytes is one of the promising and applicable approaches to increase the maturation and quality of oocytes used in in vitro fertilisation (IVF). On the other hand, oocyte vitrification is another approaches to help couples, especially in females who have cancer and need to undergo chemotherapy or radiation therapy. Today, it is known that embryo incompetence for implantation is one of main causes of IVF failure, which occurs for embryos obtained from IVM and vitrified-warmed oocytes. Hence, increasing the quality of oocytes and the subsequent embryos is one of the interesting and promising field for many researchers and scientists. The conditioned medium of granulosa cells, which are closely associated with the developing female gamete, seems to be a useful approach to enhance the maturation of oocytes, as well as to increase the quality of embryos. In this study, we investigated the quality of embryos obtained from fresh and mature vitrified-warmed oocytes in granulosa cell-condition medium (GCCM).
  • Methods: For this purpose, the fresh and vitrified mouse germinal vesicle oocytes were kept in GCCM to get matured (MII stage). Then, the mature oocytes were transferred to IVF medium and fertilized with adult NMRI mouse sperms.
  • Results: The rate of embryo formation from fresh and vitrified-warmed mature oocytes was evaluated. Our analysis showed that the GCCM reduces the deleterious effects of freezing step and increases the rate of embryo formation, as well as improves the quality of the resulting embryos.
  • Conclusion: These data suggest that GCCM might be used as IVM medium for human oocytes and increase the rate and quality of embryos, as well as raise the rate of implantation and pregnancy.
