• MiR-199a-5p and 3120-3p suppress cell survival, proliferation and angiogenesis by inhibiting Ras, IKK, NFkB1, PI3K in colorectal and gastric cancer

  • Introduction: Dysregulation of microRNA expression, plays a major role in the development and progression of most human malignancies including colorectal and gastric cancer. MicroRNA, regulates gene expression by inducing RNA regulation or interfering with translation miRNA and their aberrant expression might be use as diagnostic and prognostic biomarker of cancer. So we purpose to evaluate the role of Ras, IKK, NFkB1, PI3K by mir-199a-5p, 3120-3p in colorectal and gastric cancer.
  • Methods: The feature of microRNA was archived by using mirbase. Valid target genes and predict one were identified by mirtarbase and mirwalk 2.0. Gene expression in normal and tumor tissue was obtained from NCBI. Finally, the pathway enrichment analysis was performed by the KEGG and David. GENEMANIA used to find gene network.
  • Results: The result dedicated that mir-199a-5p and 3120-3p by inhibiting Ras which actives Raf1, MEK, ERK through phosphorylation prevent proliferation of cancer cell and angiogenesis. IKK, NFkB were inhibited by blocking PI3K, which actives Ras. So MYC, BCL through inhibition cell survival, suppress cancer. Mir-199a-5p, and 3120-3p by inhibiting Ras, PI3K, IKK, NFkB through preventing cell cycle, prevent cancer development. Mentioned microRNAs, block NFkB by inhibiting PI3K which actives Act. So NFkB by inhibiting VEGF, suppresses cancer through inhibition angiogenesis.
  • Conclusion: Mir-199a-5p, 3120-3p by effecting on Ras, IKK, NFkB1, PI3K, in PI3K-Act, RAS, TNF and colorectal pathway effect on proliferation, angiogenesis and cell survival. Mentioned microRNAs by acting as tumor suppressor prevent cancer development and tumor spread by inhibiting proliferation of cancer cell, angiogenesis and sell survival.
