• Comparison of antitumor effects of mono and bis-benzimidazole : A systematic review

  • Introduction: Cancer is characterized by the wild growth of abnormal cell .WHO expected to be 12million deaths per year by 2030. Benzimidazole and derivatives exhibit pharmacological activities such as anticancer. Mono and bis benzimidazole have shown strong binding intraction with DNA. The aim of this systematic review is comparing the effects of mono and bis benzimidazole in the growth inhibitory of cancer cells for optimum use in the pharmaceutical sciences.
  • Methods: Search was done by keywords in the databases: Pubmed, Google scholar, Science Direct, in the period of 2000-2019. At the pubmed, 10 articles were received. By studying the title of the articles, 6 articles were selected. By studing the absteract of the articles, 4 articles were selected. At the Google Scholar,12 articles were obtained ,5 articles were included . At Science Direct, articles were in English.4 review articles were identifiend and all removed. Finally we review 5 articles in this study.
  • Results: Five experimental studies were reviewed. A comparative study of 46 cancer cell lines showed that mono benzimidazole derivatives had higher cytotoxic activity than bisbenzimidazole. Actually , bisbenzimidazole moiety is more favorable for cytotoxic and mono benzimidazole for cytostatic activity. Two other studies found the activity of monobenzimidazole to be more efficient because it binds to DNA and interacts more with cancer cell proteins. In one comparative study with a three-phase in vitro assay, monobenzimidazole showed a stronger binding to the DNA of cancer cells than bisbenzimidazole. A study of 60 cell lines showed that mono benzimidazole had higher anticancer activity than bis benzimidazole. In fact, the link between DNA binding and anticancer activity has been demonstrated.
  • Conclusion: Most articles have reported the more effective of monobenzimidazole on antitumor properties, growth inhibition and antiproliferation. The use of mono benzimidazole derivatives in drug production for reduces cancer and accelerates the healing process is effective.
