  • Introduction: Sodium nitrite is widely used in meat products such as sausages and kielbasa as preservatives and preserves the sharp taste of these compounds. Recent studies have shown that this compound can cause carcinogenesis by causing nitros amine production in the body and can be harmful and deadly to cells. On the other hand, essential elements of the body such as zinc, copper, and selenium are involved in the structure of many body enzymes and proteins such as cytochromes, glutathione peroxidases, aldolases,..., and thus play important roles in the survival of cells. The purpose of this study was to consider the effect of three essential elements of zinc, copper and selenium on the viability of normal fibroblasts cells of skin that have been largely degraded by sodium nitrite.
  • Methods: The combination of sodium nitrite with 5 concentrations (25, 50, 125, 250 and 500 μg / ml) was prepared and each concentration was exposed to skin fibroblast cells with 3 replications. The MTT method was used to evaluate their effect on cells. The ELISA reader was used at 570 nm to evaluate the cell status. Then Cu, Zn and Se were each exposed to 5 concentrations (10, 50, 125, 250 and 500 μg / ml) were prepared and each cell line was exposed to these elements at different concentrations (each with 3 replications) and their effect on cells was evaluated by MTT method. Finally, ELISA Reader was used for final cell evaluation
  • Results: The results showed that sodium nitrite combination significantly decreased the growth of normal fibroblast cells with IC50 = 230/5636 μg / ml. Exposing these cells to zinc, copper, and selenium significantly increased the growth of these cells. The highest mean cell viability was observed at concentrations of 500 μg / ml each
  • Conclusion: The results of this study demonstrate the essential role of zinc, copper, and selenium in enhancing the viability of normal dermal fibroblasts as well as protective factors against the deleterious effects of sodium nitrite.
