• Evaluation of the miRNA-34C therapeuticeffects on the programed cell death in colorectal cancer cell line

  • Introduction: MiRNAs (micro RNAs) are small intracellular RNAs , that has many effects on the expression of genes that often reduce the translation of mRNA into the desired protein. MiRNAs regulated cancer target genes and therefore they use as therapeutic agent for cancer treatment.
  • Methods: SW480 , colon cancer cell line, treated with miRNA34-C , Scrambled andlipofectaminein three groups. apoptotic changes were examined byannexin-PI . total RNA was extracted from using TRiZoL .cDNA was synthesizes using a cDNA synthesis kit (TAKARA-japan) and the level expression of Caspase3,Caspase7 and Caspase9 were examined in parallel with beta-actin (as housekeeping genes) by using Real Time PCRtechnique in different groups.
  • Results: The expression levels of defined genes and poptosis induction were significantly increased in the miRNA34-C treated cells compared to the scrambled sequence and Lipofectamin groups. ectopic expression of miRNA34-C can lead to increased expression level of Caspase3, Caspase7 and Caspase9 genes and induce apoptosis pathways. there fore in this study suggested, miRNA34-C may be used as therapeutic agent for inducing apoptosis in colon cancer cells.
  • Conclusion: there fore in this study suggested, miRNA34-C may be used as therapeutic agent for inducing apoptosis in colon cancer cells.
