• Evaluation of antioxidant compounds and inhibitory effects of Apium graveolens extracts on xanthine oxidase activity

  • Introduction: The Reactive Oxygen Species (ROS) damage to cells led to the developmental and progression of many disease such as cancer, gout, aging, diabetes and cardiovascular disease. Gout is a chronic metabolic disease leads raise uric acid levels in the blood by purine degradation. The most effective enzyme in creating gout is xanthine oxidase which can produce a lot of superoxide anion through the consumption of oxygen. Thus recently, the plant as natural sources of antioxidant are used in protect against oxidative damage, as an ingredient for cosmetics, drugs and food. The purpose of this study was to determine the medical importance of Apium graveolens through investigating their inhibitory effects of the crude extract on xanthine oxidase besides their content and activity of some antioxidant compounds.
  • Methods: To do this research, Apium graveolens harvested, they dried in dark and powdered. Xanthine oxidase inhibitory effect of extracts assayed. Also, total phenol, flavonoid, and anthocyanin contents of plant sample determined and their correlation with xanthin oxidase inhibitory effect was defined, too.
  • Results: The results showed that the extract of Apium graveolens exhibited a good inhibition effects on xanthine oxidase activity and the most inhibitive effect observed at the level of 0.3 mg/ ml with 5.82 %. Total phenol, flavonoid and anthocyanin contents were 7.78±0.195 mgEG/g dw, 0. 825± 0.051 mgEQ/g dw, 3.25± 0.229 mg/g dw, respectably. Also, the effect of total phenol and flavonoid on xanthin oxidase inhibition showed an efficient correlation (p˂ 0.05).
  • Conclusion: Therefore, these results suggest that the extract of Apium graveolens is a rich source of antioxidant and an effective inhibitor of xanthin oxidase enzyme and based on that, it is recommended for pharmacological studies.
