Apoptotic and cytotoxic effect of the thiosemicarbazone complex with zn2+ in k562 cell line

Abstract


Introduction

thiosemicarbazone family is organic compounds with antitumor, antibacterial and antiviral effects. in the present study, we have investigated cytotoxic and apoptosis induction of the new thiosemicarbazone complex with zn2+ in k562 cell line.

Methods

the cytotoxicity of the compound was evaluated via mtt assay. morphological evaluation of the apoptotic cells were done using the fluorescent acridine orange and ethidium bromide dyes and then were observed with fluorescence microscopy.

Results

The data demonstrated the growth inhibitory effect of the compound in k562 cells. the inhibitory concentration 50% (ic50) was calculated 100 µm in the cells after 72 h-incubation with the compound. according to results, an induction of apoptosis was initiated by the compound in the k562 cells (at ic50 value) after 24-72 h. the apoptotic cells exhibited bright green color with condensed and fragmented dna.

Conclusion

According to these data, we can conclude that thiosemicarbazone complex with zn2+ has potential role as a therapeutic factor to induction of apoptosis and can be considered for its further development as an antileukemia drug.
