Dysregulation of mir-485-3p expression in advanced stage colorectal cancer tissues ; a pilot study on 10 iranian patients

Abstract


Introduction

Colorectal cancer (crc) , is the third most common cancer type and the leading cause of cancer death worldwide. currently , researches on micrornas and their roles in cancer progression gain remarkable attention . micrornas ( mirnas ) , are a family of small and highly conserved ncrnas with 20-25 nucleotides in length, could regulate genes expressions by binding to 3ˊ-untranslated regions (3ˊutr) of target mrnas and participate in many biological processes including cell proliferation, differentiation and apoptosis. moreover, emerging evidences has demonstrated that abnormal expression of mirnas was involved in cancer progression so they could be recognized as oncogenes or tumor suppressors. mir-485-3p , is located at 101055419-101055491[+] genomic region on the chromosome 14. also , it could function in mitochondrial respiratory, migration , invasion and metastasis. in this study , we clarified that mir-485-3p expression was downregulated significantly in advanced stage colorectal cancer tissues compared to matched non-cancerous ones.

Methods

In this study, 10 paires of advanced stage crc fresh tissues samples and their adjacent normal ones were obtained from iran national tumor bank , kept frozen in liquid nitrogen and stored in -80 centigrade until rna extraction . total rna was extracted by using rnx-plus solution .cdna was synthetized by takara kit and the expression of mir-485-3p in tumor and adjacent normal tissues were determined by real-time pcr.

Results

The results of our study showed that the expression of mir-485-3p was decreased notably in cancer tissues compared to matched normal tissues.

Conclusion

Collectively, this study revealed that mir-485-3p was extensively downregulated in advanced stage tumor tissues compared to matched adjacent normal ones and can act as a tumor-suppressor gene in advanced stage colorectal cancer. besides, mir-485-3p can consider as a therapeutic and putative biomarker in colorectal cancer.
