Abstract


Introduction

introduction: colorectal cancer is the third common cancer in women and men and mostly be derived from adenoma polyps that include tubular, villous and tubulo-villous types. mmp-7 gene plays a critical role in angiogenesis, growth and malignancy conversion in colorectal adenoma polyps. investigation of some genes which play role in growth and angiogenesis can be used as a molecular diagnosis for adenoma polyp progress to malignancy. the aim of this study is investigation of mmp-7 gene expression in different types of colorectal adenoma polyps and malignancy incidence.

Methods

Materials and methods: in this descriptive – analytical study during 2015 to 2016, 50 biopsy samples of adenoma polyp and 20 normal samples were collected from participations of colorectal cancer screening program in taleghani hospital.the clinical information of patients was collected by a questionnaire. mrna was extracted by yekta tajhizkit and cdna was synthesized by takara kit. mmp-7 gene expression was investigated by real time pcr method and relative quantification. fold change of gene expression was evaluated by (2-∆∆ct) method. data was analyzed by 7500system sds version 1.3 and prism version 5 applications.

Results

result: among 50 adenoma polyp samples, 14 (28%) villous adenoma polyps, 19(38%) tubular adenoma polyps and 17 (34%) tubulo-villous adenoma polyps were detected. mmp-7 mrna expression was significantly higher in adenoma polyps compared to the control group (22).

Conclusion

conclusion: generally, villous type has more malignancy potential. in the present study, mmp-7 gene exhibited high expression level in villous adenoma polyps, it may be a reason for malignancy progressing. our data showed that mmp-7 gene can be used as a therapeutic target for cancer prevention.
