Investigating the relationship between polymorphism of p22phox in smoker population with chronic obstructive pulmonary disease (copd) and susceptibility to disease

Abstract


Introduction

Copd (chronic obstructive pulmonary disease) is a slowly progressive condition characterized by airflow limitation, which is largely irreversible. inflammatory responses are the major hallmark of copd. the oxidative stress is the hallmark of copd involving several candidates. one of them, the nad(p)h oxidase mediates increased oxidative stress through increased release of superoxide anion. the enzyme is comprised of several distinct subunits and one of the electron transfer subunits, p22phox, expressed in vascular smooth muscle cells, is a critical component for ros generation. p22 phox is a polymorphic subunit of nad(p)h-oxidase that is critical for activation and stabilization of the enzyme. this enzyme is involved in the production of superoxide that triggers inflammatory injuries to the copd.

Methods

100 healthy subjects (control) and 213 patients with copd were selected for this study. the c242t polymorphism of p22phox was determined using polymerase chain reaction-amplification refractory mutation system (pcr-arms). according to p22 genotype, the subjects were divided in wild-type (cc) and t allele carriers (ct + tt). the oxidative stress markers were selected for the study. the measurements were made on a high-throughput spectramax plus 384 spectrophotometer.

Results

There is significant relationship between p22 phox gene polymorphisms and disease. the relationship between the number of exacerbations in patients with copd and polymorphism of p22 phox showed no significant difference between the number of exacerbations in patients with copd and p22 phox polymorphism. there was no significant association between p22phox c242t polymorphism and disease severity in patients with copd. there is a significant relationship between genotype of p22 phox and tac (or= 0.06 (95% ci: - 0.11_-0.00); p= 0.03) and there were no significant relationship between genotype of this group and clinical characteristics and the other oxidative stress parameters.

Conclusion

These results suggest that the p22phox c242t polymorphism may be a major risk factor for copd. moreover, the results of plasma antioxidant capacity in smokers and patients dropped by increased attacks.
