Down regulation of bcl2 and up regulation of bax during 2-ndc mediated apoptosis in acute promyelocytic leukemia nb4 cells

Abstract


Introduction

Dithiocarbamates have been known as important chemical synthetic compounds for treatment of cancer.

Methods

: in the present study, we have evaluated growth inhibition and induction of apoptosis in nb4 cells by 2-nitro-1- phenylethylpiperidine-1- carbodithioate (2-ndc) from dithio-carbamate family. the nb4 cells were cultured in 96 wells plate and treated in 10-120 μm of the compound for 24, 48 and 72 hours. the percent of cell viability and growth inhibition were assessed by mtt assay. for evaluation of bax and bcl2 expressions, we treated nb4 cells with ic50 concentration of 2-ndc during 24, 48 and 72 hours. then rna was extracted from the cells by a special procedure and cdna was synthesized for each sample. finally the percentage of gene expression was measured by real time pcr.

Results

: 2-ndc inhibited cell growth and viability with ic50 of 20 μm. results showed that 2-ndc increased the expression of bax as a proapoptotic gene and decreased bcl2 (anti apoptotic gene) expression, in the treated nb4 cells.

Conclusion

Based on these data, we confirmed that 2-ndc can be a good candidate for further evaluations of treatment for leukemia.
