Evaluation of antioxidant activity and cytotoxicity of an ethanol extract of leaves and fruits of mespilus germanica on human lung cancer a549 cell line

Abstract


Introduction

Lung cancer is the most common cancers and natural products extracted from medicinal plants can play an important role in cancer treatment.

Methods

In this study to determine the content and activity of some antioxidant compounds of its leaves and fruits and the inhibitory effects of their crude extract on lung cancer a549 cell line. therefore, leaves and fruits of mespilus germanica were harvested from the tonekabon city. then, the cells exposed to different concentrations of m. germanica (31.25 to 2000 μg/ml) and were incubed for 24, 48, and 72 hours and mtt assay was used to determine cytotoxicity. also, total phenol, flavonoid, anthocyanin, and carotenoid contents were determined as well as antioxidant activity of extract using dpph method.

Results

The results showed that the highest total phenol, flavonoids, anthocyanins, and carotenoids in leaves were more than those in fruits. further, its different concentrations effected on dpph radical inhibition as the highest inhibitory effect were 72.1 % and 85.5 % recording at the concentrations of 1.8 mg/ml respectively extracted from its leaves and fruits. the results have also shown that cancer cell growth was inhibited by different concentration of leave and fruit after 24, 48, and 72 hours and the most pronounced growth inhibition recorded was 90.38 % in 2000 μg/ml of the fruit extraction.

Conclusion

These results suggest that the leaves and fruits of mespilus germanica have the most antioxidant activity and cytotoxicity against a549 cell line. it seems to come with further research and utilizes its compound in cancer treatment.
