Introduction: water-soluble acrylamide (acr) monomer is used in different industries. acr is found in carbohydrate – rich food cooked at high temperatures. acr monomer is a potent neurotoxic and damages the central and the peripheral nervous system in human and animals. different mechanisms are involved in acr-induced neurotoxicity. in recent years, oxidative stress and apoptosis have been mentioned as important pathways in acr neurotoxicity.morin is an active and important compound of moraceae (mulberrywhite) in different studies morin has shown high antioxidant and anti-apoptotic activity.therefore this study was designed to investigate the possible protective effect of morin on acr-induced neurotoxicity in in vitro (pc12 cells) model.
For in vitro assay, pc12 cells were cultured and exposed to morin (25, 50,100,200, and 400 μg/m) for 24 h. then, acr at final concentration of 4/5 mm was added to each well. mtt test was performed to assay cell viability. apoptotic cells (the percentage of cells in sub- g1 population) were evaluated by pi staining. accumulation of ros was measured using the dcfh-da flow cytomery method.
The exposure of pc12 cells to acr reduced cell viability, increased sub g1 population. results showed that acr increased ros in cells and ros played an important role in acr cytotoxicity. also, morin inhibited ros generation in cells exposed to acr.
Oxidative stress and apoptosis played an important role in acr- induced neurotoxicity. morin administration significantly reduced acr neurotoxicity. morin decreased acr- induced apoptosis
