Optimization of the best concentration of aloe vera extract for treating gastric cancer cells in order to reduce her-2 gene expression

Abstract


Introduction

Her2 is a glycoprotein that has tyrosine kinase activity and plays an important role in guiding growth signals. its expression is multiplied in most cancer cells. in gastric cancer, this gene sometimes has a 60-fold increase in expression. the aim of this study was to evaluate the expression of her2 gene in gastric adenocarcinoma cells after treatment with aloe vera extracts. this plant, with its anti-cancer properties, inhibits tumor formation in cancer cells.

Methods

105ags cells were cultured in 12 wells plates.the cells were treated with 800, 1200 and 2000 μg / ml of aqueous extract of aloe vera for 48 hours. the rna extraction andcdna synthesis have done using special kits. finally, her2 gene expression was evaluated with specific primers and real time pcr. gapdh gene was used as an internal control gene.

Results

Analysis of the results have shown that in concentrations of 800, 1200 and 2000μg / ml, we observed a 4, 1.3 and 0 fold decrease in expression of her2 gene, respectively.

Conclusion

According to the results, the most reduction of her-2 gene expression, have seen in concentration of 800 μg / ml aloe vera. it seems that this herbal extract has the most effect in low concentration. herbal medicines are a good alternative to chemical drugs, given the effectiveness of preventing tumor growth and fewer side effects. in this study, the her2 gene, as one of the factors in tumor malignancy, has been reduced significantly by affecting the aloe vera extract.
