Evaluation of adenovirus in pseudomonas aeruginosa susceptible keratitis by pcr molecular method

Abstract


Introduction

Keratitis is inflammation of the cornea of the eye. in keratitis, microbial agents are important. in the presence of infectious keratitis can be mentioned in bacteria, fungi and viruses. adenovirus is usually associated with eye diseases, respiratory system, gastroenteritis, heart disease, neurological diseases, and genitourinary diseases. eye keratitis is one of the major problems that caused by adenovirus in people. considering the importance of this bacterium in infectious diseases, especially ocular diseases in this study is attempted to investigate the role and rapid identification of this virus in suspected pseudomonas keratitis. in fact, the main issue of this study is the molecular identification of adenovirus in pseudomonas aeruginosa keratitis infections by pcr method

Methods

In this study, 70 samples of pseudomonas aeruginosa suspected keratitis were collected by a specialist physician from lababinejad hospital. pcr test of adenovirus was optimized and performed from specificity and lod test. final optimized pcr test was done in addition to positive and negative controls.

Results

Pcr product with size of 301 bp was observed in 1.5% gel agarose. in specificity test all of microorganism’s dna were negative except pseudomonas aeruginosa’ dna. from 70 samples of pseudomonas aeruginosa suspected keratitis 3 samples were positive for adenovirus

Conclusion

Particularly in this study, it has been proven that part of suspected pseudomonas keratitis is due to adenoviruses. in this regard, pcr test can be used as a quick, precise and sensitive method for diagnosis.
